You may be a bit short on details.

Does the game start? If not, make sure you have windows media player activated.

Does it crash while running (and possibly starting)?

Look in:

... your folder ....\Documents\My Games\Distant Worlds Universe

And see if there is a crash file.

If you used the matrix installer, make sure you say "yes" to anything it asks about installing.

You can manually update .NET framework - maybe there's something borked with it on your PC. (google installing latest version of .net)

99% it's always Windows Media Player, no one use it :) And game is not work without it.

Weird. It looks like it is not even trying. Does the button to open the manual work?

You have never had a successful run? Which would rule out any mod issues causing this...

I would try to remove and reinstall the game.

As Bingeling say, copy content of file here: C:\Users\[user name]\Documents\My Games\Distant Worlds Universe\DW_CrashDump.txt

It's XNA can't load graphics drivers from DirectX so reinstall both from \Redist folder in game directory. DirectX and XNA 4.0 (fix, remove and reinstall) or upgrade online to XNA 4.5.2 already newest.
