I would like to know what the effect of an AGC is on assault landings? Does it need to be carrying supplies to work?

Lesser disruption/casualties, higher AV.
Does it need to be carrying supplies to work?
No, but it MUST carry an Amphibious Force HQ. These units will only load onto AGCs, and should never be unloaded. Also, don't confuse them with Amphibious Command HQs, which are simple Corps HQs.
